Nematodes Sample Collection
Sampling Nematodes in Soil:

The samples must be collected by a person specially trained for this job. It is preferable that it is always the same person.

The sample must be taken with a hole or tube open on one side, manufactured for this purpose.

The sampling must be carried out in the root zone between two drippers (nematodes flee from areas of soil saturated with water), at a depth of 25 cm.

At the site where the sample is taken, the first 5 cm of the soil surface should be removed (nematodes flee from areas of dry land).

A sample is made up of 12 to 15 sub-samples, which must be collected in the area where the presence of nematodes is suspected. The sub-samples taken are placed in a bucket and mixed carefully. From this mixture, 1 kg is taken, which constitutes the sample to be shipped to the laboratory.

Each sample must be identified with the corresponding data: name of the farm, contact person, telephone, e-mail, date of sample collection, crop, lot / block and type of analysis required.

Exposure to the sun and high temperatures should be avoided (nematodes die at a temperature greater than 48º C).

Root nematodes sampling:

The samples must be collected by a person specially trained for this job, it is preferable that it is always the same person.

Samples should be collected from plants where a population of phytophagous nematodes is suspected.

The sample must be composed of 10 - 15 plants (= subsamples). To take root samples, a 20-30cm hole should be dug next to the plant. With scissors, a small amount of roots should be cut (10 to 20 grams per plant), which should not have a diameter greater than a pencil (0.5 cm). Roots, tertiary or secondary roots can be used.

The roots of the 10 to 15 plants (subsamples) are collected and placed in a plastic bag. Each sample must be identified with the corresponding information: name of the farm, contact person, telephone, e-mail, date of sample collection, crop, lot / block and type of analysis required.

The sample should be shipped to the laboratory within two days from the collection of the sample. Exposure to the sun and high temperatures should be avoided (nematodes die at a temperature greater than 48º C).
